摘要
通过观察鲤鱼精子的泳动情况,研究了DNA大分子(约含1×105b)及其经超声波处理后的较小分子片段(约含6×103b)钠盐溶液对鲤鱼精子活力的影响.结果显示,在相同的盐浓度下,精子在片段化的较小分子DNA溶液中存活时间随DNA浓度升高而相应缩短,当DNA浓度为50μg/mL时,其存活时间最长,达20min;在50~150μg/mL的DNA大分子钠盐溶液内精子的存活时间相近;而当DNA浓度为200μg/mL时,精子的存活时间则明显缩短.此外,DNA分子大小对精子的存活时间没有明显影响.然而,DNA钠盐溶液内盐的浓度则与精子存活时间有密切关系,精子在0.4%NaCl溶液中存活时间最长.
Based on the observation of the carp sperm swin,a study of the effect of sodium salt solution of DNA mac molecules (about 100 kb) and ultrasonic fractured DNA fragments (about 6 kb) on its activity was made.The result showed that the survival time of the carp sperm in DNA fragments solution decreased gradually as the DNA concentration increased under the condition of the same sodium salt solution.When DNA concentration was 50 μg/mL,the sperm survived up to 20 min,the longest survival time.When in the sodium salt solution of DNA macro molecules at the concentration ranging from 50 μg/mL to 150 μg/mL,the sperm survived the similarly long, and when the concentration was up to 200 μg/mL,the survival time significantly decreased.No matter whether DNA was of macromolecules or micromolecules,it had no significant effect on the survival time of the sperm.The relationship of DNA solution concentration with the survival time of the sperm was a bell like curve in diagram.The sperm survied the longest when in the solution of 0.4% NaCl.
