摘要 目的:建立测定烟草愈伤组织中有效成分茄尼醇的反相高效液相色谱分析方法,并用该方法测定不同激素组合、不同时间培养的烟草愈伤组织。方法:以 Symmetry C_(18)(150mm×4.6mm,5.0μm)为色谱柱;甲醇-无水乙醇(1:1.5)为流动相,流速为1.5mL·min^(-1);枪测波长为215nm,灵敏度为2.00AUFS。结果:茄尼醇浓度在0.05~1.10mg·mL^(-1)(r=0.9997)范围内线性关系良好,平均加样回收率(n=6)为98.6%,RSD=1.3%。在一定的激素组合条件、一定时间的培养下茄尼醇的含量有规律地变化,其中在附加激素2,4-二氯苯氧乙酸(1mg·L^(-1))的 MS 同体培养条件培养40d 时烟草愈伤组织中的茄尼醇的百分含量达到0.882%。结论:该 RP-HPLC 法是一种快速、简便、准确的分析方法,为探索烟草组织培养最佳条件提供了重要手段。 Objective: To establish a sensitive RP - HPLC method for determining the content of solanesol in tobacco callus cultivated at different plant hormones combination and different culturing time. Method:Making use of Symmetry C18 ( 150 mm × 4.6 mm,5.0 μm) as column, and in the ratio ( 1 : 1.5 ) with methanol and anhydrous etha- nol,the flow rate was 1.5 mL · min^-1 ,with wavelength at 215 nm and sensitive degree was 2.00 AUFS. Result: The linear range for solanesol would be good within 0.05 - 1.10 mg· mL^ - 1 ( r = 0. 9997 ), the average recoveries ( n =6) were 98.6%, RSD = 1.3%. Under certain plant hormone s combinations and time period, the content of solanesol would have changed in disciplinary way, especially when the callus cultivated on the regulator MS with 2, 4 - dichlorophenoxy acetic acid ( 1 mg · L^ - 1 ) as solid culture for 40 days, the content of solanesol in the callus may reach 0. 882%. Conclusion:The method is accurate, reliable and suitable for determination of the content solanesol in tobacco callus and the other tobacco tissue culture.
