摘要
目的研究三七总皂甙对大鼠脑出血后核转录因子κB(NF-κB)表达的影响。方法将SD大鼠随机分为正常组、假手术组、脑出血模型组、三七总皂甙治疗组,采用VII型胶原酶注入大鼠右侧苍白球制备脑出血模型,每组分别在术后6、24、48、96h4个时点收取脑组织标本,免疫组化检测NF-κBp65蛋白表达,同时测量脑组织含水量。结果正常组和假手术组比较,大鼠脑出血后脑组织NF-κBp65表达以及脑组织含水量显著增高(P<0·01);与脑出血模型组比较,三七总皂甙治疗组NF-κBp65水平(P<0·01)及脑组织含水量(6hP<0·05,24、48、96hP<0·01)均显著降低。结论三七总皂甙可以抑制脑出血后NF-κB表达,这可能是三七总皂甙减轻脑出血后脑水肿的机制之一。
Objective To study the influence of sanqizongzaodai on the expression of NF-κB in the perihematoma regions of rats brain. Methods Sixty-four Sprague Dawley male rats were randomly divided into normal group (A), pseudo-operation group (B), model group (C) and sanqizongzaodai treatment group (D). ICH models of rats were induced by injection of V Ⅱtype collagenase into the right globas pallidus. The brain tissue specimen in each group were gathered at 6,24,48and 96 h after operation, respectively. The expression of NF-κB P65 in rat brain after ICH was detected with immunohistochemistry method,and at the same time the moisure in brain tissue was measured. Results Compared with group A and group B , the expression of NF-κBP65 and moisture in brain tissue in group C were obviously increased ( P〈0. 01). Compared with group C , the expression of NF-κB P65 and moisture in brain tissue in group D were obviously decreased. Conclusion Sanqizongzaodai can reduce the expression of NF -κB in rat after ICH, which may be related with its anti-edema effect following ICH.
