摘要
目的:探讨枸杞多糖对新西兰兔动脉粥样硬化形成的作用及机制。方法:将40只雄性新西兰兔随机分为4组,每组10只,对照组喂普通饲料;模型组,普通饲料加质量分数为1.5%胆固醇喂养;枸杞多糖(LBP)组,普通饲料加质量分数为1.5%胆固醇喂养,同时按体重腹腔注射LBP(3.0mg/kg);卡托普利组,普通饲料加质量分数为1.5%胆固醇喂养,同时按体重灌喂卡托普利(15.0 mg/kg)。10周后,测定各组空腹血清三脂酰甘油(TG)、总胆固醇(TCh)、高密度脂蛋白胆固醇(HDL-C)、一氧化氮(NO)、内皮素-1(ET-1)、丙二醛(MDA)、C反应蛋白(CRP)含量以及超氧化物歧化酶(SOD)活性,计算主动脉内膜粥样斑块面积并观察主动脉形态学变化。结果:模型组与对照组比较,TG、TCh、NO、MDA、ET-1、CRP明显升高(P<0.01),HDL-C/TCh、SOD活性明显下降(P<0.01),主动脉内膜粥样斑块面积较大;而LBP组与模型组比较,TG、CRP、NO、ET-1、MDA明显下降(P<0.01),TCh无明显变化,HDL-C/TCh比值、SOD活性明显升高(P<0.01),主动脉内膜粥样斑块面积明显减少(P<0.01),模型组与卡托普利组各项指标无差异。结论:枸杞多糖对动脉粥样硬化有明显的预防和治疗作用,其机制可能与抗氧化、调节免疫、提高HDL-C/TCh有关。
Objective: To investigate the mechanism of Lycium Barbarum Polysaccharide on the formation of atherosclerosis in New Zealand rabbits. Methods: 40 healthy New Zealand white rabbits were randomly divided into four groups, the control group was fed with normal diet for 10 weeks, the model group was fed with normal diet plus 1.5% cholesterol, the LBP group was fed with normal diet plus 1.5% cholesterol and 3.0mg/kg LBP, the Captopril group was fed with normal diet plus 1.5% cholesterol and 15.0 mg/kg Captopril. After ten weeks'experiment, each group's triglyceride, total cholesterol , high density lipoprotein cholesterol, C reactive protein, nitric oxide , maleic dialdehyde and endothelin 1 were measured, so was the activity of superoxide dismutase. The area of atherosclerosis mottle in tunica intima of aorta was computed and the morphological change of aorta was observed. Results" Compared with the control group, model groups' TG,TCh, NO, MDA, CRP and ET-1 increased significantly( P 〈 0.01), activity of SOD and HDL-C/TCh remarkably decreased(P 〈 0.01). While compared with the model group, the LBP group's TG, TCh, NO, MDA, CRP and ET-1 significantly lowered( P 〈 0.01), the activity of SOD and HDL-C/TCh remarkably increased( P 〈 0.01). Compared with model group, Captopril group did not have significant change. Conclusion: LBP can effectively accentuate the activity of SOD against oxidation, can regulate immunity, and can elevate the radio of HDL-C/TCh, to protect endothelial function against atherosclerosis, so LBP is a potential drug in treating and preventing atherosclerosis.
