β-胡萝卜素脂质体对大鼠中性粒细胞内外源活性氧损伤膜的保护作用 被引量:1

Protective Effects of β-Carotene Liposome against Rat Neutrophile Membrane Damage by Intra-or Extra-Cellular Reactive Oxygen Species

摘要 用DPH和N-(3芘)马来酰亚胺荧光标记物标记大鼠中性粒细胞膜脂及膜蛋白质,测定荧光参数及变化以探讨β-胡萝卜素脂质体对细胞内外源活性氧致大鼠中性粒细胞膜损伤的保护作用。结果表明β-胡萝卜素脂质体对细胞外源活性氧造成的膜脂、膜蛋白质的损伤及内源活性氧造成的膜蛋白质损伤均有较好的防护效果,对细胞内源活性氧造成的膜脂损伤的防护效果有待进一步探讨。 In this experiment the protective effects of β-carotene(β-C)liposome against rat neu-trophile membrane damage caused by extracellular or intracellular reactive oxygen werespecies(ROS)investigated by means of fluorescence labels 1,6-diphenyL-1,3,5 hexatriane(DPH)and N-(3-pyrene)maleimide(N-(3p)M).The extracellular ROS include the singletoxygen(1O2)generated by photoreaction of sensitizer rosebengal and the superoxide anionproduced by xanthine/xanthine oxidase reactive system.The intracellular ROS wassenshizer rose6engal and superoxide anion()produced by xanthine/xanthine oxidase reac-tive system while the intracellular oxygen was generated by stimulating neutrophile respira-tory burst with opsonized zymosan. No change in the peak or shape of fluorescent excitationand emission spectra was found for either of the two labels.The results of fluorescent polar-ization value in the n1embrane showed that β-C liposome could significently inhibit the dam-age of rat neutrophile membrane protein reacted either by intracellular generated reactiveoxygen species or by extracellular generatedO2 or1O2.It could also inhibit the injury of therat neutrophile membrane lipid reacted by extracellular reactive oxygen species.However,the protective effects ofβ-C liposome against membrane lipid damage reacted by intracellularreactive oxygen species by at the san1e concentration as those on membrane protein were notsignificent, which could be partly explained by relatively higher damage level of rat neu-trophile membrane lipid domain reacted by intracellular reactive oxygen species than that ofthe membrane protein domain.
