摘要
采用正交实验探讨了微波辅助提取柿叶多糖的最佳条件,研究了醇析浓度对多糖得率的影响;用苯酚-硫酸显色法测定了最佳醇析浓度和最佳提取条件下的柿叶多糖含量。结果表明:最佳提取条件为,料液比(m:v)=1:40、微波功率450W、提取时间6min,最佳提取条件重现性良好;最佳醇析浓度为90%;柿叶干粉粗多糖得率为14.8%,粗多糖中多糖含量为90.8%,柿叶干粉多糖含量为13.4%;平均加样回收率为98.2%,相对标准偏差RSD=1.39%(n=5)。
Microwave-assisted extraction technique was used for the extraction of polysaccharides from D.kaki L. and the extraction conditions were optimized by orthogonal experiment in this work. Also, study the influence of ethyl alcohol concentration in water on yield of polysaccharides. The results indicated that the best extraction was carried out under 450 W microwave power for 6 min using 40 mL extraction solvent per gram sample powder. The yield of polysaccharides obtained was 14.8 % and the polysaccharides puritlty in produced product was 90.8 %. The recovery obtained was 98.2 % and the R.S.D. of analytical method was 1.39 %.
