摘要
目的:观察BDNF和TrkB在唾液腺腺样囊性癌组织和细胞系中的表达,揭示BDNF和TrkB与唾液腺腺样囊性癌高转移特性和神经侵袭特性的关系。方法:以30例SACC标本为研究对象,以5例正常腮腺及3例腺泡细胞癌作为对照,采用免疫组化法对BDNF和TrkB进行检测。以SACC高、低转移细胞系ACC-2和ACC-M为研究对象,利用免疫组化、RT-PCR、实时荧光定量PCR和Western印迹,检测BDNF和TrkB在其中的表达差异。采用χ2检验进行统计学分析。结果:组织标本免疫组化显示:TrkB表达水平在嗜神经现象组显著高于未见嗜神经现象组(Ρ<0.05),BDNF表达水平与嗜神经现象无关(Ρ>0.05)。实时荧光定量PCR和Western印迹均检测到BDNF和TrkB在ACC-M中的表达高于ACC-2(Ρ<0.01)。结论:TrkB的表达强度与SACC的神经侵袭特性相关,BDNF和TrkB的表达强度与SACC的高转移特性相关。
PURPOSE: To investigate the expression of BDNF (brain derived neurotrophin factor) and its receptor TrkB (tyrosine kinase B)in SACC (salivary adenoid cystic carcinoma) tissue and cell lines and to evaluate the relationship between the expression of BDNF and TrkB and the perineural invasion and metastasis characteristics of SACC. METHODS: BDNF and TrkB expression were examined in 30 cases of SACC, 10 normal parotid glands and 10 acinic cell carcinoma tissue samples by immunohistochemical staining. BDNF and TrkB were proved expressed in both low metastasis SACC cell line (ACC-2) and high metastasis SACC cell line (ACC-M) by immunohistochemical staining, RT- PCR, Real-time PCR and Western blot. x2test was used for statistical analysis. RESULTS: TrkB expression in the group with perineural invasion was significantly higher than the group without perineural invasion (P〈O.05). RT-PCR, Real-time PCR and Western blot confirmed that BDNF and TrkB expressed in both ACC-2 and ACC-M. BDNF and TrkB expression in group of ACC-M was significantly higher than that of ACC-2 (P〈0.01). CONCLUSION: Enhanced expression of TrkB may contribute to perineural invasion in SACC and enhanced expression of BDNF and TrkB may contribute to the metastasis of SACC.
