17-β雌二醇对高糖环境中内皮细胞MT1-MMP mRNA表达变化及其凋亡的影响 被引量:2

摘要 目的:研究雌二醇(E2)对高糖(Glu,30mmol/L)环境中内皮细胞系EVC-304的凋亡及其膜型基质金属蛋白酶-1(MT1-MMP)mRNA表达的影响.方法:用E2干预高糖环境中的ECV-304,用四唑盐比色试验(MTT)检测高糖和E2对ECV-304增殖的影响;用流式细胞术(flowcytometry,FCM)和细胞基因组DNALadder法检测高糖和E2干预后ECV-304凋亡;用半定量RT—PCR法检测高糖和E2干预后ECV-304MT1-MMPmRNA表达.结果:高糖可以促进EVC-304凋亡(P〈0.01),也可以降低MT1-MMP的表达(P〈0.01);E2在浓度为0.01~0.10mmol/L时,可以拮抗高糖的促凋亡效应(P〈0.01),也可恢复性上调MT1-MMP的表达(P〈0.05).E2的这种效应不随时间变化而变化(P〈0.05).结论:高糖对内皮细胞具有毒性作用,E2对内皮细胞具有保护作用;E2还可以上调高糖环境中内皮细胞MT1-MMP的表达. AIM: To explore the effect of 17-β estradiol ( E2 )on apoptosis and membrane-type matrix metalloproteinase 1 ( MT1- MMP) mRNA expression in endothelial cell line EVC-304 under the high glucose condition 30 mmoL/L. METHODS: The proliferation of EVC-304, which is treated by high glucose(30 mmoL/L) and/or E, of different concentrations respectively, was measured by MTI" assays. Then the apoptosis of ECV-304 was detected by flow cytometry ( FCM ) and DNA ladder assays. Finally, MT1 - MMP mRNA expression was measured by semi-quantitative RTPCR. RESULTS: After ECV-304 had been treated by high glucose(30 mmol/L) , its proliferation and mRNA expression of MT1-MMP was inhibited significantly( P 〈 0.01 ). However, the tendency fall back after E2 had been administrated ( P 〈 0.01 ). Furthermore, this effect of E2 reached a peak at its concentration from 0.01 to 0. 10 mmoL/L. The apoptosis was only detected in cells which had been administrated by high glucose and E2 at low (0.001 nmol/L) and high ( 1.0 nmol/L) concentration by DNA Ladder assays. CONCLUSION: High glucose has a strong toxicity on vascular endothelial cells. Moderate level of E2 has a protective effect on vascular endothelial cells. E2 can enhance MT1- MMP expression of endothelial cells under high glucose condition.
