摘要 [目的]通过对酶联免疫吸附试验(ELISA)双抗原夹心法艾滋病病毒抗体检测中实验条件的改进,在确保检测结果的前提下,探索出操作性更强的实验方法。[方法]以国际公认的实验条件设立对照组,设立不同实验条件的实验组,找出和标准对照组没有统计学差异的实验组。[结果]温度均为37℃,结合时间为50min,显色时间为20min的实验组和标准对照组总体均数无统计学差异,且消耗的总时间最短。[结论]温度均为37℃,结合时间为50min,显色时间为20min的实验组可以作为传统方法的替代方法加以推广。 Objective To obtain novel and convenient method that substitute the standard method in detection of HIV antibody in antigen based Sandwich ELISA assay. Methods A novel method for HIV detection through modifying experimental conditions was estabhshod, and method was selected that provided results without any significant difference compared with those obtained by standard assay. Results Our data showed that binding time for 50 minutes at 37℃, followed by color reaction for 20 minutes, the result did not change significantly compared with standard assay. However, our method remarkedly shortens detection duration. Conclusions The novel method with binding time for 50 minutes at 37℃, color reaction for 20 minutes could substitute the standard method in clinical diagnosis of HIV.
