摘要
目的从细胞水平阐明骨折愈合过程中软骨痂向硬骨痂转变演化过程。方法采用小鼠胫骨不稳定骨折模型,对小鼠的骨痂进行X线摄片,HE染色,B rdu掺入检测,ColⅡ、ColX的原位杂交检测,Tunel法对原位凋亡检测。结果骨折修复愈合中后期软骨细胞大量而有序的凋亡,协同邻近编织骨内大量细胞增生侵入才是其主要的演变过程。结论肥大软骨细胞的有序凋亡是干预骨折愈合过程的潜在环节。
Objective To clarify the replacement view of cellular level during fracture healing. Methods process of cartilaginous callus by callous callus from the With the instablized fracture models of tibia of mice,the callus were examined by X-ray pictures, hematoxylin-eosin( HE ) staining, In situ hybridization of Col Ⅱ, Col Ⅹ, Brdu incorporation assay,TdT-mediated dUTP nick end-labeling( TUNEL) assay. The changes of cartilaginous callus was detected. Results Regular apoptosis of hypertrophic chondrocytes cooperating with mass proliferation of cells in neighboring woven bone is the primary replacement process during fracture healing. Conclusion Regular apoptosis of hypertrophic chondrocytes is the potential step by intervention in fracture healing.
