细胞因子HGF体外增强骨肉瘤细胞株的增殖和黏附性 被引量:2

Increased Proliferation Rate and Adhesiveness of Osteosarcoma Cell Lines by HGF Cytokine in Vitro

摘要 【目的】研究肝细胞生长因子/离散因子(HGF/SF)及其受体蛋白c-Met在骨肉瘤细胞株MG-63和HOS中的表达及对瘤细胞生物学行为的影响,探讨HGF/c-Met系统在骨肉瘤细胞生长侵袭过程中的作用。【方法】用流式细胞术检测HGF-!、HGF-"、c-Met在2株骨肉瘤细胞株中的表达;外源性HGF刺激此2株细胞株后,用MTT法、黏附性检测方法研究细胞增殖速率和黏附性的变化。【结果】HGF-!、HGF-"、c-Met在2株骨肉瘤细胞株中的阳性表达率均低于3%。HGF浓度达50ng/mL以上时,MG-63和HOS的增殖速率显著增高,P<0.01。当外源性HGF的浓度分别达5ng/mL和10ng/mL时,MG-63和HOS的黏附性指标即显著升高,P<0.01。外源性HGF刺激后HGF-!、HGF-"、c-Met在2株骨肉瘤细胞株中的表达略有变化,但差异无统计学意义,P>0.05。【结论】HGF可以促进骨肉瘤细胞株MG-63和HOS的体外增殖和黏附能力。 [Objective] To investigate the expression of hepatocyte growth factor/scatter factor (HGF/SF) and its receptor protein c-Met in osteosarcoma cell line MG-63 and HOS, and the influence of HGF on the proliferative and adhesive abilities of the two osteosarcoma cell lines, so as to investigate the possible role of this cytokine during the process of osteosarcoma proliferation and invasion. [Methods] Two osteosarcoma cell lines MG-63 and HOS were traditional cultured. Flow cytometry analysis was used to detect the positive rates of HGF-α,HGF-β,c-Met in the two cell lines. Then, MTT method and cell adhesion detection were used to detect the change of the cells' proliferation rate and adhesive abilities after being stimulated by external pure HGF. [Results] The positive rates of HGF-α,HGF-β,c-Met in the two cell lines were all lower than 3%. The proliferation rates of MG-63 and HOS increased dramatically when the concentration of external HGF reached 50 ng/mL. Only 5 ng/mL external HGF could make the index of MG-63 adhesion increase markedly (P〈 0.01). Similarly, 10 ng/mL external HGF could make the index of HOS adhesion increase significantly(P〈 0.01). That the percentage of HGF-α, HGF-β, c-Met positive cells almost had no variation after stimulated by external HGF. [Conclusions] HGF cytokine could enhance the proliferative and adhesive abilities of osteosarcoma cell lines MG-63 and HOS in vitro.
