右归丸对实验性甲状腺功能减退大鼠骨骼肌GluT4 mRNA表达水平的影响 被引量:10

摘要 目的:研究温阳补肾中药右归丸对实验性甲状腺功能减退症大鼠的治疗作用及对骨骼肌葡萄糖转运蛋白4(GluT4)mRNA表达的影响。方法:采用丙基硫氧嘧啶灌胃给药造成实验性甲减大鼠模型,然后给予不同的治疗方案,治疗8周后处死大鼠,取血检测甲状腺功能,取肌肉组织检测GluT4mRNA表达量。结果:模型组大鼠骨骼肌GluT4mRNA表达显著降低,约相当于正常表达水平的14.37%。治疗后右归丸中剂量组、右归丸高剂量组、中西药合用组GluT4mRNA表达明显提高,大致恢复正常范围;而西药组、右归丸低剂量组、自拟方组与模型组相比无明显变化。结论:右归丸能上调甲减大鼠骨骼肌GluT4mRNA表达水平,促进骨骼肌对葡萄糖的转运,从而改善骨骼肌细胞的能量代谢,这可能是中药能有效治疗甲状腺功能减退症的作用机理之一。 To study the effect of Yougui Wan on experimental hypothyroidism and GluT4 mRNA expression in the skeletal muscle of experimental hypothyroidism rats. Methods: Experimental hypothyroidism rat model was made with intragastrical perfusion of propylthiouracil(PUT) and was treated with different treatment programs. After treatment for 8 weeks.the rats were killed and the blood sample was taken for examination of thyroid hormones,and the muscular tissue was taken to detect GIuT t mRNA expression. Results: GluT4 mRNA expression in the skeletal muscle significantly decreased in the model group. corresponding to 14. 37% of the normal expression; After treatment,the GluT4 mRNA expression significantly increased to about the normal range in the Yougui Wan mid dose group, Yougui Wan high dose group,and the integrated Chinese and western medicine group. There was no significant changes as the western medicine group, Yougui Wan low dose group, self-stipulated recipe group compared with the model group. Conclusion:Yougui Wan can up-regulate GluT4 mRNA expression in the skeletal muscle of hypothyroidism rats,promoting transport of glucose in the skeletal muscle,so as to improve energy metabolism of the skeletal muscular cells,which is possibly one of the mechanisms of the Chinese medicine effectively treating hypothyroidism.
