选择性超深低温和复苏对猴脑缺血的影响

摘要 目的观察选择性超深低温和复苏对猴脑缺血的保护作用。方法5至6岁健康恒河猴4只,在灌注前先阻断双侧颈总动脉血流10min,然后建立闭胸式脑局部体外循环,行一侧颈内动脉冷灌注,同侧颈内静脉回流。60min后恢复脑血流,实验动物自然复苏。手术前后行头颅MRI检查,手术中持续行脑电图监护并监测血流动力学指标变化,术后行神经功能缺失评分。结果4只实验猴均成功建立实验模型,术中血流动力学稳定,术后安全复苏并长期存活,术后头颅MRI检查正常,术后脑电图正常,术后神经功能评分无异常。结论选择性超深低温和复苏对猴脑缺血有明显的保护作用。超深低温下猴脑可耐受缺血60min而无明显异常。 Objective To observe the protective effect of selective ultraprofound hypothermia and resuscitation on monkey ischemic brain. Methods For the four 5 - 6 year - aged healthy maca mulattas, bilaferal common carotid artery blood flow were blocked for ten minutes before perfusion, then brain- local extractorporeal circulation was established by right internal carotid artery ultraprofound hypothermic perfusion and homolateral internal jugular vein backflow. Brain blood flow was recovered from circulatory arrest of 60 minutes and the monkeys came back naturally. MRI examination was performed before and after operation, and the EEG and the hemodynamical parameters of the animal were observed during the experiment, then neurological deficit evaluation was performed after operation. Results The experimental model was established successfully in the four monkeys and the hemodynamical parameters were steady during the operation. All of the monkeys lived up for ever. The MRI examinations of the monkeys were normal after operation. EEG was normal and the function of neurological deficit scale was normal after operation. Conclusion There is remarkable protective effect of selective ultraprofound hypothermia and resuscitation on monkey ischemic brain. The monkey brain can tolerate ischemia for 60 min under ultraprofound hypothermia.
