摘要
未研究采用反转录多聚酶链反应技术观察了低氧对培养的大鼠大脑皮层微血管内皮细胞中诱导型一氧化氮合成酶基因表达的影响。发现经3%O2(培养液中氧分压为6.7kPa)作用24小时后,能够明显诱导内皮细胞中该型合成酶基因的表达,且在复氧12小时后,仍不能消除上述作用;而11%O2(培养液中氧分压为10.7kPa)作用相同时间,则不能诱导该酶的表达。结果表明,较严重的低氧条件可以直接诱导一氧化氮合成酶的表达,提示NO在低氧下大脑皮层微循环的调节中可能起重要作用。
Present study observed the direct effects of hypoxia on expression of inducible nitric oxide synthetase(iNOS)in endothelial cells(CMEC)which isolated from rats'cerebral cortical microvessels,using reverse transcription polymerase chain reaction(RTPCR)technique. Hypoxic mixed gases were over the cultural endothelial ce11s in different time with BNA310N2-O2-CO2 incubator(TAEAI ESPEC Co).The experimental results showed that serious hypoxia,the partial pressure of O2 is 6.7kPa in cultural medium,can induce the expression of iNOS gene in CMEC after 24 hours,reoxygenation cannot entire eliminate the effect.The moderate hypoxia, the partial pressure of O2 is 10.7 kPa, cannot induce the expression of iNOS gene,These results suggested that hypoxia could be an inducer of production of iNOS in CMEC and NO may play an importent role in regulation of cerebral microcirculation under hypoxia.
