摘要
目的 建立一种反相高效液相色谱法(RP—HPLC)检测肿瘤患者血清中甲氨喋呤(MTX)浓度的方法。方法 色谱柱为反相柱C18(150.00mm×4.60mm),柱温35℃。流动相为0.15mol/L乙酸钠:乙腈:89:11(体积比),流速1.5ml/min。进样量为10μl,检测波长303nm,用高氯酸沉淀血清标本的蛋白。结果 血清中MTX的检测范围为0.25~100.00mg/L,决定系数(r^2)=0.999,线性关系良好,平均回收率100.95%,日内及日间相对标准差(RSD)均〈5%。结论 RP—HPLC适用于临床对血药浓度的监测。
Objective To establish a reverse phase performance liquid chromatography (RP-HPLC) to detect methotrexate (MTX) level in serum of patient with tumour. Methods The RP-HPLC column was C18 reverse phase column ( 150.00 mm ×4.60 mm) and at the temperature of 35 ℃. The mobile phase was 0.15 mol/L sodium acetate : acetonitrile = 89 : 11 (V/V). The flow rate was 1.5 ml/min and 10 μl was injected in the sampler. The detective wavelength was 303 nm. The protein in serum sample was deposited down by perchloric acid. Results The linearity was shown over the detection range of MTX in serum of 0.25 - 100.00 mg/L ( r^2 =0. 9999). The average recovery was 100. 95%. The intra-day relative standard deviation (RSD) and inter-day RSD were lower than 5%. Conclusions The RP- HPLC can be used to monitor the level of MTX in serum.
