摘要
本研究通过测定24个小麦品种全麦粉中多酚氧化酶在不同酶活抑制剂作用下的酶活性变化情况,比较了几种常用酶活抑制剂巯基乙醇、EDTA、饱和NaC l和SDS(十二烷基磺酸钠)对全麦粉中多酚氧化酶活性的抑制效应,同时着重分析了巯基乙醇、EDTA和SDS在不同浓度下对多酚氧化酶的抑制效率。结果表明:不同浓度的抑制剂对同一小麦品种全麦粉的PPO活性抑制效果不同;相同浓度的抑制剂对全麦粉的PPO活性抑制效果因品种而异;在同一种浓度下(0.05mol/L)巯基乙醇和SDS对多酚氧化酶活性的抑制效果较强,其次是EDTA。用相对活力百分数证明了0.05mol/L巯基乙醇的抑制效果明显较相同浓度的EDTA和SDS两种抑制剂稳定。
Twenty -four wheat cuhivars were selected as experimental material, and polyphenoloxidase (PPO) was extracted by citric acid - phosphate buffer from their whole wheat flours. Several chemical reagents including mercaptoethanol, EDTA, saturated NaCl and SDS (sodium dodecyl sulfate) as PPO inhibitors were put into the enzyme extract, and PPO activity was determined using spectrophotometer. The results show that the inhibition effect on PPO activity is dependent on the inhibitor concentration and wheat cultivar. With inhibitor concentration of 0.05mol/ L, the inhibition efficiency of mercaptoethanol and SDS on PPO activity is relatively strong, followed by EDTA. It is proved by means of relative activity percentage that the inhibition efficiency of mercaptoethanol is more stable than EDTA and SDS in 0.05mol/L concentration.
