摘要
为了寻找可产生鬼臼毒素类化合物的内生真菌,从砂地柏(Sabina vulgarisAnt.)不同组织中分离到126株内生真菌。以鬼臼毒素,脱氧鬼臼毒素和苦鬼臼毒素为标准品,采用HPLC法筛选得到一株可代谢产生重要药用和杀虫活性物质鬼臼毒素类似物的内生真菌SC13。对菌株SC13进行ITS-5.8S rDNA区域扩增,测序及遗传进化关系分析,初步推测该菌株属于链格孢属(Alternariasp.)真菌。采用小叶碟添加法测定该菌株发酵液萃取物对三龄初期粘虫的拒食活性,其48h拒食中浓为7.259 mg.mL-1,采用浸毒叶片法测定了三龄初期小菜蛾的毒杀活性,其48h致死中浓为6.136 mg.mL-1。
126 strain endophytic fungi isolated from healthy organism of pharmaceutical plant Sabina vulgaris Ant. The culture were extracted with chloroform and analysed by HPLC. The endophtytic fungi stain SC13 could produce picropodophyllotoxin. The antifeedant activity against the 3rd larvae of Mythirnna separate Walker. were assayed with leaf-disk method and the poisionous activity against the 3rd larvae of Plutella xylostella L. were assayed with Leaf Dipped Method. The bioassay results showed that the activity of the extracting mixture of strain SC13 had some activities, the AFC50 against the 3rd larvae of Mythirnna separate Walker. during 48h was 7.23 mg · mL^-1, and LC50 against the 3rd larvae of Plutella ccylostella during 48 h was 7.23 mg · mL^-1 ITS-5. 8S rDNA sequence were used to characterize the stain SC13, and showed that the stain SC13 belong to Alternaria.
