白藜芦醇体外抑制类风湿关节炎滑膜细胞的增殖及其机制 被引量:12

Inhibitory effect of resveratrol on the proliferation of synoviocytes in rheumatoid arthritis and its mechanism in vitro

摘要 目的:检测白藜芦醇(Res)体外对类风湿关节炎(RA)滑膜细胞增殖的影响,并探讨其可能机制。方法:用不同浓度Res处理RA滑膜细胞24h后,采用四唑蓝(MTT)法检测滑膜细胞的增殖水平;用缺口末端标记法(TUNEL)和流式细胞仪检测其凋亡百分率;采用Western-blot分析caspase-3酶原的裂解;用比色法测定Res(200μmol/L)处理RA滑膜细胞不同时间及不同浓度的Res处理RA滑膜细胞12h后,caspase-3的相对活性。结果:加入不同浓度Res处理RA滑膜细胞24h后,增殖水平均显著低于对照组(P<0.01);对照组与不同浓度Res处理组凋亡细胞百分率比较,差异有统计学意义(P<0.01);用200μmol/LRes分别处理RA滑膜细胞不同时间,caspase-3活性与未处理组比较,差异有统计学意义(P<0.01);用不同浓度的Res处理细胞12h,caspase-3活性成浓度依赖性升高;用不同浓度Res处理培养的RA滑膜细胞24h,caspase-3酶原的表达随药物浓度的增加而减少。结论:Res在体外抑制类风湿关节炎滑膜细胞增殖,诱导细胞凋亡,可能与caspase-3的活化有关。 Objective To investigate the effect of resveratrol on the proliferation and apoptosis of synoviocytes in patients with rheumatoid arthritis ( RA ) in vitro and explore its mechanism. Methods The levels of cell proliferation of synoviocytes in RA after 24 h treated with. different concentrations of resveratrol were measured by monotetrazolium colourmetric assay method. The percentages of synoviocytes apoptosis in RA after 24 h treated with different concentrations of resveratrol were tested by TUNEL and flow cytometry. The relative activities of caspase-3 were determined by colorimetric assay after 4, 8, 12, 18, and 24 h treated with resveratrol (200 μmol/L) and 12 h treated with different concentrations of resveratrol. The cleavages of pro-caspase-3 were analyzed by Western blot after 24 h treated with different concentrations of resveratrol. Results The levels of cell proliferation of synoviocytes with RA after 24 h treated with different concentrations of resveratrol were significantly decreased compared with the control group ( P 〈 0.01 ). The percentages of the apoptotic cells were increased of resveratrol-treated after 24 h, the apoptosis rates between the treated groups and the control group were significantly different ( P 〈 0.01 ). When the synoviocytes in RA were treated with 200μmol/L resveratrol for different time respectively, the caspase-3 activity began to rise significantly at 4 h, reaching the peak at 12h, and was still much higher than that of the control group at 24h (P 〈 0. 01 ). After the ceils were treated with different concentrations of resveratrol for 12 h, caspase-3 activity increased in a concentration-dependent manner. After synoviocytes in RA were treated with different concentrations of resveratrol for 24 h, the expressions of pro-caspase-3 decreased as the concentration increased, the caspase-3 active μmol/L and was increased at 400μmol/L. Conclusion noviocytes and induces cell apoptosis in rheumatoid arthritis tion of caspase-3. fragment P 11 ( 11 kD ) appeared at 100 Resveratrol inhibits the proliferation of synoviocytes and induces cell apoptosis in rheumatoid arthritis in vitro, which may relate to the activa- tion of caspase-3.
