摘要 【目的】探讨微波消融对小鼠肝癌细胞活性和HSP70表达的影响。【方法】建立C57BL/6J小鼠Hepa1-6肝癌皮下移植模型,采用微波能量以不同的温度消融肝癌,分别用酶组织化学和Westernblot检测被消融肝癌组织细胞琥珀酸脱氢酶(SDH)的活性和HSP70的表达,并观察消融后30d肝癌局部残留率。【结果】经40℃、45℃或50℃,120s的条件微波消融后,50℃组消融后24h小鼠肝癌组织细胞的琥珀酸脱氢酶活性明显低于假消融组;该组消融后30d肝癌局部残留率明显低于假消融组(均P<0.01)。3个消融组消融后12h和24h肝癌组织HSP70的表达均明显高于相应的假消融组(均P<0.01),且随消融温度升高表达明显增强。【结论】微波消融可促进小鼠肝癌HSP70的表达,同时在一定消融条件下能明显降低肝癌细胞的活性。 [ Objective ] To investigate the effect of microwave ablation on the cell activity and heat shock protein 70 (HSP70) expression of liver cancer in mouse. [Methods] A C57BL/6J mouse model of liver cancer was established by subcutaneous injection of Hepa 1-6 ceils. The transplanted tumors were subjected to microwave ablation at different temperature by using microwave power, and the succinic dehydrogenase (SDH) and HSP70 expression of the ablated liver cancer were analyzed by enzymohistoehemistry and Western blot, respectively. The rate of local residual liver cancer 30 d after ablation was investigated. [ Results ] After ablation under the condition of 40 ℃, 45 ℃, or 50 ℃ for 120 s, the SDH activity of liver cancer ceils 24 h after ablation in 50 ℃ group was lower than that in sham ablation group (SMA) and the rate of local residual liver cancer 30 d after ablation in 50 ℃×120 s group was significantly lower than that in SMA group (all P〈 0.01). The HSP70 expressions of liver cancer 12 or 24 h after ablation in three ablation groups were significantly higher than that in corresponding SMA group (all P〈 0.01), and they were up-regulated in correlation with ablation temperature. [Conclusion ] Microwave ablation can promote the expression of HSP70 in transplanted liver cancer in mouse, and simultaneously, at a certain ablation condition, it can significantly decrease cell activity of liver cancer.
