摘要
应用固相抗原筛选法,建立了人抗HBsAg单克隆抗体组合文库。以液相HBsAg包被酶标板,对抗HBsAg血清阳性供者文库经多次筛选、扩增,建立起针对HBsAg的组合抗体子文库。选取12个克隆进行抗体片段表达研究,并对该抗体进行特异性酶联免疫测定,其中5株为抗-HBs阳性,应用组合文库制备人源单克隆抗体是─种简便、实用的方法。
Con the methods of antigen based selections,one human monoclonal antibody fragment library to hepatitis B surface antigeus(HBsAg)was generated. Liquid HBsAg were coated on the ELISA plate and one combinatorial antibody library that was from a HBsAg-positive donor was panned with the HBsAg coated wells,The antigen specific phages in the library were selected and enriched to form a HBsAg-specific library,Choosing 12 clones for Fab expression and anti-HBsAg specificity study,Fab supernatants from 5 clones show anti-HBs positive in enzyme linked immunoassay. Results showed that application of combinatorial library to prepare specific Fabs against viral diseases is a attractive and hopeful fields.
