摘要
采用了AtlantisC18反相色谱柱(适用于分离极性分子的反相色谱柱),成功地分离了去甲肾上腺素(NA),肾上腺素(A),多巴胺(DA)等儿茶酚胺(CAs)类物质,研究了CAs在Atlantis C18和Novle pak C18柱的保留特性,比较了流动相中离子对试剂辛烷基磺酸钠(B8)浓度对CAs容量因子k’的作用,确认采用AtlantisC18柱分离CAs,色谱条件优化空间更大,更适于复杂生物样品分析。色谱条件:Atlantis C18柱,电化检测,流动相:NaH2PO450 mmol/L、柠檬酸钠50 mmol/L缓冲液-乙腈(体积比(75∶25),检测限(S/N=2)在0.93—2.43pg,绝对回收率均在98.02%以上,样品日内RSD1.5%以下。方法精密、准确。
A method for determination of noradrenaline (NA), adrenaline (A) and dopamine (DA) using Atlantis C18 column was investigated. The retention characteristics of catecholamines (CAs) in Atlantis and Novle Pak C18 column were observed, and the contribution of defferent concentrations of octanesufonat sodium (BS) in mobile phase to capacity factor k' was compared. Experiments showed that it was better to use Atlantis C18 for separating CAs in optimizing chromatographic system, and more suitable for separating the complicated biologic samples particularly. Chromatography conditions, the mobile phase was a mixture of buffer and acetonitrile (V: V=75 : 25) and the buffer was NaH2PO450 mmol/L and citric acid 50 mmol/L. The detection limits of the method are 0. 93 to 2.48 pg(S/N=2). The recovery is above 98.02% and the relative standard deviation (RSDs) is 1.5% in a day.
