摘要 目的探讨雏鸡高度近视眼眼球后壁组织血管活性肠肽受体2亚型(vasoactive intestinal peptide receptor 2,VIPR2)mRNA的动态表达及意义。方法选择出生1 d健康来亨雏鸡共30只,15只右眼遮盖作为实验组,分别持续遮盖1周、2周和4周;另15只双眼均未遮盖作为正常对照组。两组均采用检影验光检测屈光度;眼科A超测定活体眼轴长度;对两组3个时间段眼球后壁行逆转录聚合酶链反应(reverse transcriptase polymerase chain reaction,RT-PCR)检测VIPR2 mRNA的动态表达。结果实验组由实验前远视[快速]变为高度近视眼,并随遮盖时间的延长、近视度数的加深,眼轴延长;两组眼球后壁均有VIPR2 mRNA的阳性表达;随着出生时间的延长,两组VIPR2 mRNA的表达均逐渐下降。与对照组相比,实验组VIPR2 mRNA的表达明显上调(P<0.01)。结论形觉剥夺可导致高度近视眼;高度近视眼较正常眼VIPR2 mRNA的表达明显上调。血管活性肠肽(vasoactive intestinal peptide,VIP)可能通过与VIPR2结合,参与了近视眼形成与发展的调控。 Objective To investigate, the dynamic expression and significance of the vasoactive intestinal peptide receptor 2 (VIPR2) mRNA at the posterior pole of the eye in high myopia. Methods Thirty one-day-old, healthy, yellow legborn chickens ,sere randomly chosen for the study. For 15 chickens, the experimental group, a method of form deprivation was used where the right eyes were covered continuously for 1 week, 2 weeks or 4 weeks. The other 15 chickens acted as a control group. Retinoscopy and A-scan ultrasonography were trod to measure refractive stains and axial length of the eye. The dynamic expression of VIPR2 mRNA at the posterior pole of the eye was tested with RT-PCR, Results The experimental eyes quickly changed from pre-experimental hypermetropia to high myopia during the. experimental period, and the severity of myopia and axial leith increased relative to the period of form deprivation. The expression of VIPR2 mRNA and the levels decreased over time. for both groups. The expression of VIPR2 mRNA in the experimental groups was significantly up-regulated compared to the. control group ( P 〈 0.01 ). Conclusions Form deprivation induces axial myopia. The increase, in the severity of myopia was related to the period of form deprivation. The expression of VIPR2 mRNA at the posterior pole of the eye of FDM chicken models was up-regulated significantly. VIPR2 may play an important role in the formation and development of myopia.
