大鼠海马和大脑皮质TRPA1 mRNA的表达 被引量:1

EXPRESSION OF TRPA1 mRNA IN THE HIPPOCAMPUS AND CEREBRAL CORTEX OF THE RAT

摘要 目的观察TRPA1离子通道(一种瞬间受体电位离子通道)在大鼠海马和大脑皮质的表达情况。方法提取SD成年大鼠大脑皮质、双侧海马和脊髓背根神经节(DRG,dorsalrootganglion)组织mRNA,应用RT-PCR(re-versetranscriptasePCR)技术检测TRPA1离子通道mRNA的表达;同时制作大鼠大脑和脊髓背根神经节冰冻冠状切片,采用原位杂交染色的方法用地高辛标记分子探针,检测TRPA1离子通道mRNA的表达。结果RT-PCR和原位杂交染色的结果均显示在大鼠海马和大脑皮质均存在TRPA1离子通道。结论在大鼠海马和大脑皮质均存在TRPA1离子通道。 Objective To investigate the distribution of TRPA1 (one kind of the TRP family of ion channels) channel in the hippocampus and cerebral cortex of rats. Methods RT-PCR was used to amplify the fragments of TRPA1 in the DRG (dorsal root ganglion), hippocampus and cerebral cortex of adult SD rats. In situ hybridization staining was also used to show the distribution of TRPA1 mRNA in the hippocampus and cerebral cortex of adult rat brain. Results Both RT-PCR and in situ hybridization staining showed that TRPA1 mRNA was expressed in hippocampus and cerebral cortex of the adult rat brain. Conclusion Our results suggest that there is TRPA1 mRNA in both hippocampus and cerebral cortex of the adult rat brain.
