摘要
用火炬松(Pinustaeda)成熟合子胚诱导产生的胚性愈伤组织建立了胚性细胞悬浮系。研究了细胞密度、继代时间和ABA浓度等对悬浮条件下胚性胚柄细胞团(ESM:Embryonalsuspensormass)及体细胞胚(SE:Somaticembryo)形成的影响。结果表明:当细胞密度为6×103个/ml、继代时间为2个月、ABA浓度为5mg/L时,最有利于ESM及SE发生。在悬浮培养条件下,还观察到了裂生多胚及子叶原基的分化。
Embryogenic cell suspension lines were established from embryogenic calli from mature zygotic embryos cultLlred of loblolly pine. Effects of different cell density,subcul-ture time,and ABA concentration on the formation of ESM (Embryonal suspensor mass ) and SE(Somatic embryo) were studied. The results show that optimum cultur conditions are cell density of 6 ×103, subculture time of 2 months, and ABA concentration of 5 mg/ L. The formation of cleaved polyembryony and the differentiation of primordial cotyle-dons were observed in the condition of liquid culture.
