摘要
目的观察褪黑素(MLT)对冈田酸(OA)致大鼠神经母细胞和小鼠胶质细胞瘤细胞的杂交细胞系NG108-15细胞tau蛋白过度磷酸化的影响。方法10 nmol/L OA作用于NG108-15细胞0,3,6,12及24 h,免疫细胞化学法和免疫印迹法(Western blot法)观察Ser-396位点磷酸化tau蛋白,观察MLT对细胞内Ser-396位点磷酸化tau蛋白的影响。结果在OA作用后3 h增高,6 h达到峰值,之后逐渐降低。选取OA作用6 h,低、高剂量MLT较模型组Ser-396位点磷酸化tau蛋白水平明显降低。结论MLT可对抗tau蛋白过度磷酸化,可能是其发挥抗阿尔茨海默病作用的机制之一。
Objective To investigate the effects of melatonin on okadaic acid(OA)-induced tau hy perphosphorylation in NG108-15 cell. Methods NG108-15 cells were treated with 10 nmol/L OA for 0, 3, 6, 12, or 24 h. Immunohistchemistry and Western blot test were used to determine phosphorylated tau protein. Results It was shown that a significant increase in Ser-396 phosphorylation of tau protein in OA 6 h group. OA-induced hyperphosphorylation of tau protein in NG108-15 cell could be attenuated by the pretreatment with melatonin. Conclusion Antagonistic effect of melatonin on Alzheimer's disease may be through attenuating tau protein hyperphosphorylation.
