摘要
为开展禾谷缢管蚜(Rhopalosiphum padiLinnaeus)抗性的分子生物学研究,设计简并引物并采用RT-PCR技术,克隆了长度分别为260、331、337bp的3个禾谷缢管蚜的羧酸酯酶cDNA片段,命名为Rp.est1、Rp.est2、Rp.est3;3个片段推导氨基酸残基分别为87、110、112个;同源性分析表明,Rp.est1、Rp.est2、Rp.est3之间的相似性在40%左右,表明其分别代表不同基因;Rp.est1、Rp.est2、Rp.est3与其他昆虫羧酸酯酶基因序列具有较高的相似性,其GeneBank登录号分别为AY622997、AY869713、AY869714。
Using RT-PCR technique with degenerate primers, 3 carboxylesterase gene cDNA fragment (Rp. est1, Rp. est2, Rp. est3) were cloned from Rhopalosiphum padi (Linnaeus), which were 260 bp, 331 bp,337 bp nucleotides in length, and encodes 87, 110, 112 amino acids, respectively. Sequence analysis showed that the 3 cloned carboxylesterase gene cDNA fragments show near 40% homology each other,and have high homology with those from other insects. The GeneBank accession numbers of the 3 carboxylesterase genes are AY622997, AY869713, AY869714.
