摘要 目的了解海南人群中人类切除修复交叉互补基因1(ERCC1)的-4533G/A多态性情况。方法用盐提取法提取人群中白细胞的DNA,以聚合酶链反应、限制性内切核酸酶检测-4533G/A多态性。结果在分析的200例海南人群中,ERCC1-4533多态性位点有GG、GA、AA3种基因型,频率分别为0.03、0.96、0.01,G和A的等位基因频率为0.51和0.49。结论ERCC1是人类核甘酸切除修复(NER)系统的重要组成部分,其表达水平与卵巢肿瘤、消化道肿瘤的耐药性有关,因此ERCC1-4533多态性的研究对于肿瘤耐药性具有重要的意义。 Objective To investigate the polymorphism of human excision repair cross - complementing gene 1 - 4533G/A in Hainan Province. Methods DNAs were extracted from the white blood cells of people in Hainan by salt - out method. Polymerase chain reaction and restriction endonuclease was used to determine the 4533G/A polymorphism. Results Three genotypes of GG, GA and AA were identified in - 4533G/A polymorphism of 200 Hainan individuals, the genotype frequencies of GG, GA and AA were 0.095,0.89 and 0.015, respectively and the allelic frequencies G and A were 0.54 and 0.46. Conclusion ERCCI is the important part of human nucleotide excision repair system and the expression of ERCCI is associated with drug resistance of ovary tumor patients and the alimentary tract tumors. It is of important significance in the study of the ploymorphism of ERCCI - 4533G/A in drug resistance of ovary and the alimentary tract tumors patients.
