摘要
通过软琼脂克隆法及有限稀释法,从(Hca-F25/L)细胞系中分离出五株不同转移力的克隆细胞,它们的共同特点是:均沿淋巴系统转移,仅有极少数(1.88%)同时伴有肺转移;五株克隆及其亲代对角蛋白单抗(HK2)均呈现阳性,提示尽管它们分化很低,但仍保持了上皮组织的特性;经染色体分析证实,每一克隆内同一核型的细胞占绝大多数,它们和亲代有相同的四个标记染色体,表明它们之间有“血缘”关系。它们的不同之处表现在:在615小鼠体内均能致瘤(100%),但在淋巴结中的转移率及其转移程度有很大的差别,其中一株高转移细胞系(78.6%—89.4%),三株中转移细胞系(33.3%—66.6%),一株低转移细胞系(15%~15.4%);五株克隆及其亲代的干系核型也各不相同;在细胞形态上和细胞电泳率方面,高、低转移细胞系与亲代细胞系三者之间有明显的差别。本文较确切地证明了 Hca 瘤株在转移表型和核型上的异质性。同时,也补充和发展了 H22/Hca 肿瘤系统。
By means of soft agar clonization and limited diluting methods,5 cell clone with different metastatic capacity have been separated from HcaF25/L cell line.Their common charaters are:All clone metastasize
along lymphatic system except a small percent (1.88%) associated with lung metastasis;5 cell clone as well as the parental generation all present keratin monoclonal antibody (HK2) positive reaction,indicate epithelial character with low differentiation;karyotype assay reveal each clone has highly identical karyotype,though they are different
from each other but they have simillar marker chromosomes (M_1,M_2,M_3,M_4).The different among them are:various lymphatic metastatic capacity,say,one highly metastatic clone (78.6—89.4%),three moderately metastatic clone (33.3—66.6%)and one low metastatic clone(15—15.4%);the three types cell clone has various morphology and different cell electrophoretic rate.All the results indicated that Hca strain is heterogeneous in phenotype and karyotype.
