摘要
在用聚合酶链反应(PCR)技术进行基因扩增研究过程中,采用酚/氯仿常规方法从人血痕中抽提DNA,所抽提DNA数量不足用于PCR扩增。为提高模板DNA的数量,尽量避免抽提过程所用试剂对PCR的干扰,先后采用裂解法、水煮法抽提DNA。结果表明,此两种方法要比酚/氯仿法简便、迅速,效果好。而在后两种方法比较下,水煮法要比裂解法更具优越性,是一种从血痕中提取DNA的良好方法。
During the study of gene amplification ,the quantity of DNA,which was extmcted from the bloodstrain of human In order to increase the quantity of templet DNA,to avoid the distmction of the chemical reagent to PCR during the procedure of chemical extraction, two methods,i.e. lysis and water-boiled,were used.The results show that these two methods are simpler, quicker and more econoli cal than phenol/chlorom methed. The effect Of the two metheds is better than the old one. Comparing the latter two methods,i.e. lysis and water-boiled, water-boiled method is a good way of extracting NDA from bloodstains, it is more advantageous than lysis methed.
