摘要
用安捷伦Zorbax SB-C18(4.6cm×250cm,5μm)色谱柱,以K2HPO4(0.02mol/L,pH值3)和甲醇(二者体积比为90:10)为流动相,在紫外检测波长218nm的条件下,利用反相高效液相色谱法(RP-HPLC)测定发酵液中庚二酸含量,获得的庚二酸吸收峰面积与质量浓度的线性相关系数为0.999。庚二酸加标回收率98.38%.101.84%,相对标准偏差为0.45%-1.0%。该方法能准确测定发酵液中庚二酸含量,而且操作简便、快捷、结果可靠。
A method for determining pimelic acid in the broth system by reversed-phase high performance liquid chromatography(RP-HPLC) was developed.The HPLC condition were as follows:Aglilent Zorbax SB-C18(4.6 cm×250cm,5μm) with a mixture of methanol(10%)and 0.02 mol/L, pH 3.0 K2HPO4 [ 90 % ( V/V ) ] as the mobile phase; UV detection wavelength is at 218 nm. The linear range of pimelic acid was 0. 999, The recoveries were 98.38% - 101.84% with relative standard deviation 0.45% - 1.0% .The method is simple ,rapid and accurate for determining pimelic acid in broth system.
