蜂胶-银离子复合纳米乳剂的研制及其杀菌活性评价 被引量:9

DEVELOPMENT OF PROPOLIS-SILVER ION COMPOUND NANOMETER EMULSION AND EVALUATION OF ITS GERMICIDAL ACTIVITY

摘要 复合纳米消毒乳剂是以蜂胶和银离子为主要抗菌成分,为了解其理化性能、杀菌效果及安全性,采用悬液定量杀菌试验和动物实验及仪器分析方法进行了实验室研究。结果,该复合纳米消毒乳剂含蜂胶20g/L,总黄酮含量为5.61g/L,银离子含量为50μg/L。透射电镜观察可见复合乳剂中颗粒直径在30~100nm,分布均匀。以含体积分数6.25%的复合纳米消毒乳剂对大肠杆菌、金黄色葡萄球菌、绿脓杆菌作用1min,杀灭率达到100%;以含体积分数12.5%该消毒乳液对白色念珠菌作用5min,杀灭率亦达100%。该纳米消毒乳液小鼠急性经口毒性LD50〉5000mg/kg,属实际无毒;该乳剂原液对家兔皮肤、眼结膜和阴道粘膜刺激指数均为0,属于无刺激性。结论,复合纳米消毒乳剂具有良好的杀灭细菌繁殖体和酵母菌的作用,动物试验结果属于无毒类物质、对皮肤粘膜无刺激。 Compound nanometer disinfection emulsion contains propolis and silver ion as the main anfimierobie ingredients. In order to know its physicochemical properties, germicidal efficacy and safety, suspension quantitative germicidal test, animal experiment and instrumental analysis were used to carry out laboratory study. Results: The compound nanometer disinfection emulsion contained propolis 20 g/L, total flavone 5.61 g/L and silver ion 50 μg/L. Transmission electron-microscopic observation found that the diameters of the particles in the compound emulsion were 30~100 nm and the particles were homogeneously distributed. 6.25% (v/v) compound nanometer disinfection emulsion with a 1 min contact time killed 100% of Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa. 12.5% (v/v) compound disinfection emulsion with a 5 min contact time killed 100% of Candida albicans. The mouse acute oral LD50 of the nanometer disinfection emulsion was 〉 5000 mg/kg which meant that it is practically non - toxic. The irritation index of its stock emulsion to rabbit skin, conjunctiva and vaginal mucosa was 0 which meant that it is non - irritating. Conclusions: The compound nanometer disinfection emulsion had good efficacy in killing vegetative forms of bacteria and yeasts. The results of animal tests indicated that it belonged to non - toxic substance and did not irritate skin and mucons membrane.
