血管平滑肌细胞受电场作用后迁移的变化 被引量:1

摘要 目的:观察生理电场干预对大鼠主动脉血管平滑肌细胞迁移的影响。方法:实验于2002-07/2003-12在解放军第三军医大学西南医院中心实验室进行。取Wistar大鼠47只,颈离断法处死大鼠,体外培养主动脉血管平滑肌细胞。将细胞根据培养基分为DMEM组和DMEM+体积分数为0.1的胎牛血清组,分别给予0,50,100,150,200,250mV/mm生理电场干预6h,显微成像及图像分析技术记录分析血管平滑肌细胞迁移。结果:①在DMEM组,细胞在250mV/mm电场中没有定向迁移,电场干预没有明显提高细胞迁移速度。②在DMEM+体积分数为0.1的胎牛血清组,血管平滑肌细胞在电场作用下显示明显的趋化反应。在100~250mV/mm,细胞向阴极迁移。在150mV/mm电场中,单个细胞和细胞团定向迁移的速度没有明显差异[(12.79±0.77),(14.21±0.45)μm/h,P>0.05]。结论:生理电场诱导血管平滑肌细胞向阴极定向迁移,电场的作用依赖于电场强度和血清成分。 AIM: To investigate the effect of physiological electric field on the migration of cultured vascular smooth muscle cells (VSMC) of arterial aorta of rats. METHODS: The experiment was eonducted at the Central Laboratory, Southwest Hospital, Third Military Medical University of Chinesl PLA from July 2002 to December 2003. A total of 47 Wistar rats were selected and killed by breaking their necks. VSMC of rats was cultured in vitro. The cells were assigned into DMEM group and DMEM+fetal bovine serum with the 0.1 volume fraction group, and then were exposed to phys!ologieal electric fields of 0, 50, 100, 150, 200 and 250 mV/mm for 6 hours. The migration of VSMC was recorded and analyzed with micro-imaging andimage analysis. RESULTS: ①In DMEM group, there was no oriented migration in 250 mV/mm electric field. The intervention of electric field did not significantly elevate the speed of migration. ②ln the DMEM+fetal bovine serum with the 0.1 volume fraction group, there was significant chemotactic response of VSMC in the effect of electric field. The cells migrated toward negative pole in 100-250 mY/ram. In 150 mV/mm, there was insignificant difference in migration speed of single cell and cell mass [( 12.79±0.77), ( 14.21 ±0.45 )μm/h ,P〉0.05]. CONCLUSION: Physiological electric field orients VSMC migration toward negative pole. The migration effect of electric field is dependent on strength of electric field and serum.
