摘要
目的:观察重组人生长激素(rhgh)对体外成骨细胞(OB)细胞周期及细胞骨架Factin的影响.方法:用浓度为10和200μg/L的重组人生长激素加入大鼠OB培养体系,观察对大鼠OB的细胞周期及细胞骨架的影响.结果:定量分析表明,在rhgh作用下,成骨细胞S期、G2/M期细胞百分比增加,OB的绿色荧光强度、红色荧光强度无显著变化.Factin纤维呈束状平行排列,荧光染色强,分布均匀.结论:重组人生长激素对OB的合成代谢有直接影响,但对细胞骨架无显著影响.
AIM: To observe the changes of the cell cycle and cytoskeleton of rat calvarial osteoblasts after stimulated by recombinant human growth hormone (rh-gh). METHODS: Osteoblastic cells from rats were incubated in the medium supplemented wiht rh-gh at different doses ( 10, 200μg/L), and then the changes of cytoskeleton and cell cycle were detected. RESULTS: Quantitative analysis showed that osteoblasts were arrested in S phase and G2/M phase, however, green fluorescent density and red fluorescent density (nuclei) were not significantly changed. The filamentous F-actin distributed in bunches and was stained strongly and evenly. The outline of nucleus was clear. CONCLUSION: Rh-gh exerts a direct anabolic effect on osteoblasts, but no obvious effect on cytoskeleton of rat osteoblasts.
