摘要
目的:研究黄酮类化合物—槲皮素、木樨草素、芹菜素、大豆苷元、染料木素、木樨草苷、忍冬苷、黄芩苷及芦丁在Caco-2细胞模型中的吸收特性,探讨其吸收与结构的关系。方法:分别测定Caco-2细胞在各种条件下对9种黄酮类化合物的吸收。结果:Caco-2细胞对9种黄酮类化合物吸收均以偏酸性介质为佳;吸收量均随着浓度的增加而呈线性增加;当pH5·5时,5种黄酮苷元的吸收量顺序为:槲皮素>木樨草素>染料木素>芹菜素>大豆苷元;4种黄酮苷的吸收量的顺序为:木樨草苷>忍冬苷>黄芩苷>芦丁。结论:①酸性介质有利于9种黄酮类化合物在Caco-2细胞模型中的吸收,介质pH的改变对槲皮素在Caco-2细胞模型上吸收的影响最大,而对芹菜素、染料木素和大豆苷元的影响较小。②9种黄酮类化合物在Caco-2细胞模型上的吸收均为被动扩散。③黄酮苷元母核上的-OH数目及B-环的连接位置均对黄酮苷元的吸收有影响,而苷元糖苷化后,其连接糖的数目、种类及连接位置在黄酮苷的吸收中起主导作用。本实验中黄酮苷元的吸收均高于黄酮苷。
AIM: To investigate uptake characteristics of quercetin, luteolin, apigenin, genistein, daidzein, luteolin-7-O-β-D- galactoside, lonicerin, baicalin and rutin in the Caco-2 cell culture model system, and the relationship between uptake and structures of flavonoids. METHOD: The uptake of nine flavonoids were determined at different medium pH and different concentrations. RESULTS: The uptake of 9 flavonoids were the best at acidity medium; and the uptake all linearly increased with the increasing of sample concentrations; In pH5.5 medium the uptake of the 5 flavone aglycone is: quercetin 〉 luteolin 〉 genistein 〉 apigenin 〉 daidzein; The uptake of the 4 flavone glycoside is: luteohn-7-O-βD-galactoside 〉 lonicerin 〉 baicalin 〉 rutin. CONCLUSION: ①Acidic medium is propitions to uptake of 9 flavonoids in Caco-2 cell culture model. The effect of pH on the uptake of Quercetin was the greatest, while Apigenin, Genistein, Daidzein smaller.②The nine flavonoids in Caco-2 cell culture model are absorbed through passive diffusion. ③The numbers of -OH and the location of B-ring affect the uptake of aglycone; The number, type and location of sugar play an important role in uptake of glycoside. This study also shows that the uptake of aglycone is much higher than that of glycoside.
