摘要
目的探讨雷公藤内酯醇(T ri)对急性实验性变态反应性脑脊髓炎(EAE)大鼠外周血CD4+、CD8+的影响。方法模型组采用豚鼠髓鞘蛋白和福氏完全佐剂诱发大鼠EAE;治疗组在模型组的基础上予以不同剂量T ri,观察各组的发病情况;流式细胞仪检测外周血CD4+、CD8+;HE染色和Loyez氏髓鞘染色分别观察病理和髓鞘改变。结果高剂量T ri组未出现临床症状,与模型组和低剂量T ri组比较,高剂量T ri组CD8+明显增高、CD4+/CD8+值明显降低(P<0.05,<0.05);脑和脊髓炎症细胞浸润明显减少;髓鞘结构完整。结论T ri对EAE的治疗有剂量相关性,其作用机制与其增加CD8+、降低CD4+/CD8+值,从而调节机体免疫平衡有关。
[Objective] To observe the effect of Triptolide (Tri) on CD4^+ and CD8^+ in experimental allergic encephalomyelitis (EAE) in Wistar rats. [Methods] The animal model was established in Wistar rats by immunization with myelin basic protein (MBP) of guinea pig spinal cord and complete Freund's adjuvant. At the same time and after immunization, Tri was administered with various dose in treatment group. The symptom of EAE was observed and CD4^+, CD8^+ of peripheral blood were detected with flow cytometric analysis, pathological feature and myelin of brain and spinal were detected with HE stain and Loyez's stain respectively. [Results] None developed EAE in 0.4 mg/(kg ·d) Tri group, and compared with model groups and 0.2 mg/(kg · d) Tri group, CD8^+ was higher and CD4^+/CD8^+ was lower (P〈0. 05, 〈0. 05), inflammation cell which around vessels was less and myelin was not damaged in 0. 4 mg/(kg · d) Tri group. [Conclusions] Effect of Tri on EAE correlates with its ability of increasing the number of CD8^+ and decreasing CD4^+/CD8^+.
