摘要
本文采用牦牛骨粉,经4M盐酸胍提取,Sephadex G-75柱层析分离,在第二、三段收集物中含促进DNA合成的物质,定性试验证实含β_2m。将有活性的部分上CM—23柱离子交换层析分离,得三个峰,其中第二峰含促DNA合成物,证实为β_2m。SDS—PACE表明分子量在11,500～10,500之间。
Pulverized yak bone was extracted with 4. M guanidine HC1 and fractionationed by gel filtration on sephadex G 75. Several biological active components were obtained when tested for its effects on DNA synthesis in osteoblast-rich cell cultures. β2m activity was identitied by radioimmunoassay. The β2m active component was further purified by ion-exchange chromatography on carboxymethyl cellulose CM-23. Three fractions were obtained. The 2nd component (peak 2) was β2m with a molecular weight of 11,000±500.
