[目的]研究DNA彗星试验技术在检测辐照鸡肉方面的应用。[方法]新鲜鸡肉,于-20℃冷冻保存7天后,分别用0kGy、1kGy、3kGy、5kGy和7kGy不同剂量进行辐照处理,解冻后分离细胞,用彗星试验测定其DNA损伤,并分析剂量反应关系。[结果]经0kGy、1kGy、3kGy、5kGy和7kGy辐照的鸡肉细胞平均尾长和尾相分别为1.02μm、2.19μm、2.88μm、3.53μm和4.11μm以及0.38、0.75、1.05、1.60和2.03。经统计学分析,辐照与未辐照鸡肉细胞彗星电泳图尾长与尾相存在显著性差异,并存在一定的剂量反应关系。[结论]彗星试验可检测由于辐照对于鸡肉组织细胞的DNA损伤,并可作为筛选技术判断该鸡肉是否经过辐照处理。
The DNA Comet Assay is described as a rapid and inexpensive screening test to identify chicken treated by radiation. The fresh chicken was frozen at -20℃ for 7 days, with the radiation of the following dose levels respectively: 0kGy, 1kGy, 3kGy, 5kGy and 7kGy. After thawing, the irradiated chicken cell was observed with a DNA comet assay. The average tail lengths and tail moments of the chicken were 1.02μm, 2.19μm,2.88μm, 3.53μm and 4.11μm, 0.38, 0.75, 1.05, 1.60 and 2.03 respectively. The statistics analysis shows that the tail length and the taft moment shows big differences by different irradiation levels and increased with the gaining doses of irradiation. The DNA Comet Assay method can be used to detect the radiation damage of chicken and to discriminate between irradiated and non-irradiated chicken.
