玫瑰挥发油抗念珠菌活性的体外研究及两种药敏试验方法的比较 被引量:17

Study on Anti-candidal Activity of the Essential Oil from Rosa rugosa in vitro and the Difference between Macro and Micro Dilution Methods

摘要 目的探讨玫瑰挥发油的体外抗念珠菌活性,同时比较NCCLS-M27-A常量和微量液基稀释法检测结果的差异。方法先采用纸片扩散法对玫瑰挥发油抗念珠菌的活性进行初筛,再应用常量法和微量法检测其对61株临床分离念珠菌的最小抑菌浓度(M IC)与最小杀菌浓度(MFC),并以氟康唑及两性霉素B作为质控药物;同时采用棋盘微量稀释法对15株念珠菌进行玫瑰挥发油与氟康唑的体外联合抑菌实验。结果玫瑰挥发油对临床分离念珠菌均有较强的抗菌活性,两种方法测定念珠菌对玫瑰挥发油的M IC值一致率为92%,二者比较差异无显著性(P=0.18)。玫瑰挥发油与氟康唑联用对氟康唑敏感株及耐药株均表现出较好的协同相加作用,两组间分数抑菌浓度指数(FIC I)比较差异无显著性(P=0.545)。结论玫瑰挥发油对临床常见致病性念珠菌有抗菌活性;常量和微量液基稀释法具有较好的一致性;玫瑰挥发油与氟康唑联用对念珠菌在体外具有协同抑菌作用。 Objective To evaluate the anti-candidal activity of the essential oil from Rosa rugosa (EORR) and compare the results of macro and microdilution tests of NCCLS-M27 - A. Methods The anti-candidal activity of EORR was proved by disc diffusion method, then the minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (MFC) of EORR were determined in 61 clinically isolated Candida strains by macrodilution method and microdilution method, respectively. Fluconazole and amphotericin B were used as quality control. Synchronously the checkerboard microdilution method was applied to assay the effect of EORR combined with fluconazole in 15 Candida strains. Results All the test showed Candida strains was susceptible to EORR,and a high accordant rate between the two dilution methods used was obtained (92%). The effect of synergize were obviously observed without antagonism in 15 tested strains. There was no significant difference in the mean fractional inhibitory concentration index (FICI) between the fluconazole-snsceptible and the fluconazole-resistant strains ( P = 0. 545 ). Conclusion The essential oil from Rosa rugosa (EORR) has anti-Candidal activity in vitro. Discrepancies between the macro and micro dilution methods are not statistically significant. The combination of EORR with fluconazole develops a good synergitic action against Candida strains.
