骨形态发生蛋白12对肌腱和韧带损伤的修复作用 被引量:2

摘要 目的:骨形态发生蛋白12属于转化生长因子β超家族成员,能诱导间充质干细胞定向分化为成腱细胞。重组人骨形态发生蛋白12不但能使腱纤维原细胞的增殖,还能使Ⅰ型和Ⅲ型原胶原高表达,有助于肌腱的修复和愈合。回顾骨形态发生蛋白12的发展及其研究成果,并对其综述。资料来源:应用计算机检索NCBIEntrezPubmed和CNKI1965-01/2004-12骨形态发生蛋白12的相关文献,检索词:骨形态发生蛋白12(bonemorphogeneticprotein-12),损伤(repairing)。资料选择:对资料进行初审,纳入标准:①有关骨形态发生蛋白12生物学效应。②有关骨形态发生蛋白12与其家族成员比较研究。排除标准:重复研究。资料提炼:共收集42篇相关文献,排除重复或类似的同一研究,28篇符合研究要求。资料综合:①骨形态发生蛋白12的生物学作用:能诱导间充质干细胞定向分化为成腱细胞。重组人骨形态发生蛋白12不但能使腱纤维原细胞的增殖,还能使Ⅰ型和Ⅲ型原胶原高表达,有助于肌腱的修复和愈合。②与其家族其他成员比较:骨形态发生蛋白12和骨形态发生蛋白-2的信号转导途径不同;虽然骨形态发生蛋白13具有近似骨形态发生蛋白12的作用,能促进成腱细胞的增殖和Ⅰ型原胶原的基因表达,但对Ⅲ型原胶原的基因表达作用不显著。结论:成功克隆骨形态发生蛋白12基因,将其用于腱损伤的修复,具有重要临床应用价值,但目前对于骨形态发生蛋白12诱导骨髓间充质干细胞定向分化为腱细胞的机制尚未阐明,还有待进一步的研究。 OBJECTIVE: Bone morphogenetic protein-12, as one member of the transforming growth factor-β superfamily, can induce the mezenchymal stem ceils differentiating into bending ceils. Recombinant human bone morphogenetic protein-12 can increase proliferation of tendon fibroblasts and the gane expressions of procollagen type Ⅰ and type Ⅲ, which is helpful to the repairing and healing of tendon tissues. In this paper, we review and summarize the development and achievement of the bone morphogenetic protein-12. DATA SOURCES: The computer-basnd online search of NCBI Entrez Pubmed and CNKI were undertaken for literatures about the bone morphogenetic protein-12 published between January 1965 and December 2004 with the key word of "bone morphogenetic protein-12, repairing". STUDY SELECTION: The data were checked up primarily. Inclusive criteria: (1)Articles about biological effect of bone morphogenetic protein- 12; (2)Stndies on difference between bone morphoganetic protein-12 and other member in the family. Exclusive criteria: repetitive study. DATA EXTRACTION: A total of 42 relevant literatures were collected, those repetitive study or similar studies were excluded, and 28 articles met the inclusive criteria. DATA SYNTHESIS: (1)Biological effect of bone morphogenetic protein- 12: It can induce mezenchymal stem ceils differentiating into bending cells. Recombinant human bone morphogenetic protein-12 can increase proliferation of tendon fibroblasts and the gene expressions of procollagen type Ⅰ and type Ⅲ, which is helpful to the repairing and healing of tendon tissues. (2)Comparison between bone morphogenetic protein-12 and other member in the family: There are different conductive ways between bone morphogenetic protein-12 and bone morphogenetic protein-2; although bone morphogenetic protein-13 has the same biological effect to bone morphogenetic protein-12, which can increase proliferation of tendon fibroblasts and the gene expression of procollagen type Ⅰ, it has not obvious effect on the expression of procollagen type Ⅲ. CONCLUSION: As the successful clone of bone morphogenetic protein- 12, it plays an important role in repairing tendon tissues and clinical application. But the mechanism of differentiation of bone morphogenetic protein-12 into bending cells has not been clarified; therefore, it need further study.
