摘要
目的探讨载脂蛋白E(apoE)基因多态性与中国东北汉族人脑梗死的关系。方法选择中国东北汉族人140例,其中72例为脑梗死患者(脑梗死组),68例为健康体格检查者和因肛肠疾病住院的患者(对照组)。采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术,检测所有患者apoE基因多态性;一步法测定70例脑梗死患者的血脂含量。结果对照组和脑梗死组apoE基因型均以ε3ε3多见,分别为67.6%、72.2%(P>0.05)。未见ε44ε。两组ε2ε2、2ε3ε、ε2ε4、3εε4基因型频率(对照组为2.9%、13.2%、0%和16.2%,脑梗死组为2.8%、15.3%、2.8%和6.9%)和等位基因ε2、ε3、4ε频率(对照组为9.6%、82.4%和8.1%,脑梗死组为11.8%、83.3%和4.9%),差异无显著性,P>0.05。各基因型脑梗死患者血浆总胆固醇、三酰甘油、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)含量无显著差异(P>0.05)。结论在中国东北汉族人中,未发现apoE基因多态性与脑梗死相关,亦未发现apoE基因多态性与脑梗死患者血浆总胆固醇、三酰甘油、HDL-C和LDL-C之间相关。
Objective To explore the relationship between apolipoprotein E （apoE） gene polymorphism and cerebral infarction of the Han nationality in Northeast China. Methods A total of 140 subjects were selected, among them 72 were patients with cerebral infarction （ CI group）, 68 were healthy subjects or hospitalized patients with anorectal diseases （control group）, apoE polymorphisms of all patients were detected by polymerase chain reaction-restriction fragment length polymorphism （PCR-RFLP）. The plasma lipid levels in 70 patients with cerebral infarction were detected by one-step method. Results apoE ε3ε3 genotype was most common both in the control group and the CI group, they were 67.6% and 72.2% respectively （P 〉0. 05）. No ε4ε4 was found. There were no significant differences （P 〉 0. 05） between groups in genotypic frequencies of ε2ε2, ε2ε3, ε2ε4, and ε3ε4 （2.9%, 13.2%, 0%, and 16. 2% in the control group; 2. 8%, 15. 3%, 2. 8%, and 6. 9% in the CI group） and allele frequencies of ε2, ε3, and ε4 （9. 6%, 82.4%, and 8. 1% in the control group; 11.8%, 83.3%, and 4. 9% in the CI group）. There were no significant differences between the levels of plasma total cholesterol （TC）, triglyceride （TG）, high-density lipoprotein-cholesterol （HDL-C） and low density lipoprotein-cholesterol （LDL-C） in different genotypes in patients with CI （P 〉 0. 05）. Conclusion No relationship has been found between apoE polymorphisms and CI, and no relationships have been found either between apoE gene polymorphism and the plasma TC, TG, HDL-Cl, and LDL-C in patients with CI in the Han nationality in Northeast China.
