摘要
目的观察活化巨噬细胞培养上清液对原代培养的大鼠胸主动脉平滑肌细胞(VSMCs)增殖的影响,以及地塞米松的干预作用。材料与方法体外培养大鼠VSMCs并制备活化巨噬细胞培养上清液,利用四甲基偶氮唑盐(MTT)法及3H-胸腺嘧啶脱氧核苷(3H-TdR)掺入法检测不同浓度的活化巨噬细胞上清液对VSMCs增殖的效果,并用地塞米松进行干预,观察地塞米松对巨噬细胞活化及其对VSMCs增殖的影响。结果(1)活化巨噬细胞培养上清液促进VSMCs的增殖,呈浓度依赖性,随浓度增加,吸光度(OD)值及每分钟闪烁次数(cpm)值也相应增加,方差分析F值分别为15.23和23.98(P<0.01);两两比较各组间差异显著(P<0.05);(2)不同浓度地塞米松干预后OD值及cpm值随地塞米松浓度增加而降低,方差分析F值分别为63.36及13.07(P<0.01);两两比较10-11mol/L、10-9mol/L及对照组之间无统计学差异,其余各组间差异显著。结论活化巨噬细胞促进VSMCs增殖,表明炎症反应是血管内支架植入术后再狭窄的重要机制之一。地塞米松能够抑制其促增殖作用,可能在一定程度上达到预防或治疗再狭窄的作用。
Objective To observe the effect of activated Mφ supematant to the proliferation of cultured Rat thoracic aortic smooth muscle cells (SMCs) in vitro, furthermore, to investigate the antagonistic effect of dexamethasone. Materials and Meth- ods Rat thoracic aortic SMCs were harvested and activated Mφ supernatant was prepared. Cell proliferations were treatment by MTT assay and measuring 3H-TdR incorporations. Furthermore, SMCs were treated with 50% (v/v) AMS plus incremental dexamethasone. As well, cell proliferation and the DNA synthesis were assessed. Results ( 1 ) AMS could progressively promote the SMCs proliferation in a dose dependent fashion. The OD value and cpm were increased accompanied with the build up of AMS concentration. The total effect was statistically significant ( OD: F = 15.23, P 〈 0.01 ; cpm : F = 23.98, P 〈 0.01 ), there was significant difference between two groups. (2) This pro proliferating effect by AMS can be inhibited by dexamethasone. According to increasing of dexamethasone concentration, the OD value and the cpm were declined. The total effect was statistically significant (OD: F = 63.36, P 〈 0.01 ; cpm: F = 13.07,P 〈 0.01), while there were no differentiation statistically among the 10^-11mol/ L, 10^-9mol/L and the contrast group. Conclusion When The activated Mφ could promote the proliferation of SMCs, this is one of the main mechanisms of restenosis after vascular interventions especially the stent transplant. The effect of pro proliferation can be inhibited by dexamethasone, anti inflammatory therapy might be one of the approaches for the prevention and cure vascular restenosis after vascular interventions.
