巯嘌呤甲基转移酶遗传多态性对硫唑嘌呤药动学和药效学影响的研究概况

摘要 目的总结近年来巯嘌呤甲基转移酶(thiopurine methyltransferase,TPMT)遗传多态性对硫唑嘌呤药动学和药效学影响的研究概况。方法查阅近年来国内外有关文献资料,进行分析、整理和归纳。结果TPMT是硫唑嘌呤代谢过程中的重要代谢酶,TPMT活性和硫唑嘌呤活性代谢物———6-鸟嘌呤核苷酸稳态浓度成负相关;TPMT遗传多态性与硫唑嘌呤的治疗效果及毒副作用密切相关。结论TPMT活性测定和基因型分析具有潜在的临床应用价值;除TPMT缺陷外,临床用药过程中应关注导致骨髓抑制的其他因素,如药物相互作用、药物或病原体引起的骨髓抑制等。
