摘要
用PCR方法扩增了低密度脂蛋白(LDL)受体基因外显子11-内含子11-外显子12片段,长约0.8kb。利用Dynabeads固相分离单链法测定了PCR产物序列,其中有105个碱基系第一次报道,结果证明:Dynabeads固相分离单链法是一种简便、可靠、快速的PCR产物测序法。
A DNA fragment (0.8kb) from exon 11 to exon 12 of low-density lipoprotein (LDL) receptor gene was obtained by PCR. The sequencing of the fragment was carried out using Dy-nabeads solid-phase sequencing technique with modification. 105 nucleotides never reported before were determined. This method is simple, rapid and reproducible.
