高三尖杉酯碱诱导原代白血病细胞凋亡及临床意义 被引量：3

摘要 目的研究高三尖杉酯碱(HHT)对原代急性淋巴细胞白血病(ALL)和急性非淋巴细胞白血病(ANLL)细胞的促凋亡作用及临床意义。方法以20例急性白血病(AL)患者(ALL 7例,ANLL 13例)为研究对象,DNA片段原位末端标记(TUNEL)方法检测凋亡细胞。结果HHT体外处理后ANLL和ALL细胞均出现典型凋亡细胞。HHT对ALL和ANLL细胞促凋亡作用差异无显著性[TUNEL阳性率(17.34±9.60)%对(26.64±20.67)%,P>0.05]。与柔红霉素(DNR)比较,HHT对ALL细胞促凋亡作用较弱[TUNEL阳性率(17.34±9.60)%对(31.64±18.84)%,P<0.05]。联合应用HHT和阿糖胞苷(A ra-C)处理原代ALL细胞,TUNEL阳性细胞率高于单用HHT处理(P<0.001)。体外HA联合处理TUNEL阳性细胞率≥39.6%组,CR率80.0%,TUNEL阳性细胞率<39.6%组,CR率25.0%。结论HHT对原代ALL和ANLL细胞均有促凋亡作用。
