次声作用对大鼠脑谷氨酸及超微结构影响的量效特征 被引量:1

摘要 目的:观察次声作用不同天数后不同时间点大鼠脑谷氨酸以及超微结构的变化,以分析次声作用对大鼠脑谷氨酸影响的量效关系。方法:实验在解放军第四军医大学次声实验室完成,选择健康雄性SD大鼠126只,随机数字表法分为6组,对照组(n=6)和次声作用1,7,14,21,28d组,次声作用组又分为作用后1,2,3,4h4个观察时间点,每个时间点6只。次声作用组大鼠暴露于16Hz90dB的次声压力舱系统,2h/d,于作用1,7,14,21,28d后,采用免疫组织化学方法检测各组动物于次声暴露结束后1,2,3,4h等4个不同观察时间点的脑皮质谷氨酸的表达情况;透射电镜观察各组动物于次声暴露结束后2h脑皮质超微结构的变化。结果:所有动物在实验过程中无脱落,全部进入结果分析。①次声作用1,7,14d组,次声暴露结束后1,2,3,4h的脑谷氨酸免疫反应阳性细胞数和平均吸光度与对照组比较差异均无显著性意义(P>0.05);次声作用21,28d组次声暴露结束后1,2h的脑谷氨酸免疫反应阳性细胞数与对照组相比明显增加(F=8246.814,P<0.01),其平均吸光度与对照组相比也有明显增加(F=8132.912,P<0.01),作用后3,4h恢复正常,与对照组比较差异无显著性意义(P>0.05)。②对照组、次声作用7,14d组的神经元细胞形态正常。次声作用21,28d组次声暴露结束后2h可见神经元出现变性改变。结论:在次声参数16Hz90dB的条件下,一定次数的次声作用后大鼠脑皮质谷氨酸表达较对照组增多,随作用结束后时间的延长可恢复正常,超微结构也出现变性改变。 AIM: To observe the effects of infrasound at different times on the changes of glutamate (Glu) and ultrastructure of the rat pallium, and further investigate the dose-effect relationship of the effects of infrasound on Glu. METHODS: The experiment was completed in the Infrasound Laboratory of the Fourth Military Medical University of Chinese PLA. Totally 126 healthy male SD rats were divided randomly into six groups: control group and infrasound groups of 1, 7, 14, 21 and 28 days, with 6 rats in each. The infrasound groups were destined to four subgroups when the infrasound had been terminated for 1, 2, 3 and 4 hours. The rats of infrasound groups were exposed to 16 Hz 90 dB infrasound barochamber system, 2 hours daily, grouping with 1, 7, 14, 21 and 28 days. With the immunohistochemistry, the expressions of pallium Glu were detected respectively when the infrasound exposure had been terminated for 1, 2, 3 and 4 hours. Transmission electron microscope was used to observe the changes of pallium ultrastructure when the infrasound exposure had been terminated for 2 hours. RESULTS: All the rats were involved in the result analysis without any drop during the experiment.①After the infrasound exposure of 1, 2, 3 and 4 hours, there was no marked indiscrimination in the number of Glu cells which were positive for immunologic reaction and the average absorbance between control group and infrasound groups of 1, 7 and 14 days (P 〉 0.05), As for the rats exposed for 21 days or 28 days, the number of Glu cells which were positive for immunologic reaction raised up obviously after the infrasound exposure of 1 and 2 hours (F=8 246.814, P 〈 0.01), the average absorbance also elevated compared with controlled rats (F=8 132.912, P 〈 0.01). However, they revived to normal after the infrasound exposure of 3 and 4 hours, without significant difference (P 〉 0.05). ②The neurons formed normally in control group and infrasound groups of 7 and 14 days. With the rats exposed for 21 and 28 days, the changes of degeneration appeared when the infrasound exposure had been terminated for 2 hours. CONCLUSION: At the infrasound parameter of 16 Hz 90 dB, the expressions of pallium GLU increase in contrast with control group after infrasound has effected for some certain times, and come back to normal as the time extends. Meanwhile, the ultrastructure appears the denaturization.
