摘要
为了进一步研究苏丹Ⅰ、Ⅲ、Ⅳ对人胃癌细胞(SGC-7901)增殖的影响,采用流式细胞仪测定了DNA含量的变化,分析细胞周期的变化.结果表明:苏丹Ⅰ、Ⅲ、Ⅳ促进了SGC-7901细胞由G1期进入S期和G2期,表现为G1期细胞平均DNA含量下降,S期DNA含量上升,G2期DNA含量上升,细胞增殖指数(PI)增加,S期细胞指数(SPF)增加.其中苏丹Ⅲ在50μg.mL-1剂量时对SGC-7901细胞增殖影响非常显著(P<0.01),苏丹Ⅰ、Ⅳ在10μg.mL-1剂量时对SGC-7901细胞增殖影响非常显著(P<0.01).苏丹Ⅰ、Ⅲ、Ⅳ是通过影响细胞周期,从而调节SGC-7901细胞的增殖活动,促进细胞分化增殖.
Objective to study the effect of Sudan Ⅰ,Ⅲ,Ⅳ on the growth of SGC -7901. The change of DNA content is determined with Flow Cytometry, and the cell cycle is analyzed. Results show that the average content of DNA decreases in G1 phase and increases in S and G2 phase. The index of PI and SPF increase. It is indicated that Sudan Ⅰ,Ⅲ,Ⅳ accelerate the cell cycle from G1 phase to S and G2 phase. It is also shown that the effects on SGC -7901 by Sudan m in the dose of 50 μg · mL^-1 and by Sudan Ⅰ,Ⅳ in the dose of 10μg · mL^-1 is very effective (P 〈 0.01 ). It is concluded that Sudan Ⅰ,Ⅲ,Ⅳ affect SGC -7901 cell cycle to advance its differentiation and proliferation.
