白藜芦醇诱导K562细胞凋亡过程中Bcl-2蛋白水平的改变 被引量:4

摘要 目的:观察白藜芦醇对K562细胞的诱导凋亡效应,探讨其可能的作用机制。方法:应用噻唑蓝(MTT)比色法、光镜、电镜观察K562细胞凋亡;流式细胞术(FCM)测定K562细胞Bcl-2、P53蛋白水平的变化;比色法检测Caspase-3活性变化。结果:白藜芦醇可抑制K562细胞增殖,光镜和电镜下观察呈典型凋亡形态改变,Bcl-2蛋白表达明显下调,Caspase-3活性明显增强。结论:白藜芦醇可通过Bcl-2依赖性Caspase-3激活而诱导K562细胞凋亡。 AIM: To observe the apoptosis of K562 cells induced by resveratrol and to explore its possible mechanisms. METHODS: MTT assay, light microscopy and electron microscopy were used to examine apoptosis in K562 cells. Expression levels of Bcl-2 and P53 antigens were mesured by FCM. Colorimetric assay was employed to detect the activation of Caspase-3. RESULTS: Resveratrol could inhibit growth of K562 cells. Typical morphological changes of apoptosis were observed through light microscopy and electron microcopy. Expression of Bcl-2 significantly decreased after application of resveratrol. Caspase-3 could also be activated by resveratrol. CONCLUSION: Resveratrol induces apoptosis in K562 cells by activating Caspase-3 via a Bcl-2-dependent pathway.
