摘要 目的检测腺病毒介导的转化生长因子(transform ing growth factorβ1,TGF-β1)基因在兔髓核细胞中的表达及其对蛋白多糖的调节作用。方法体外分离培养兔髓核细胞,以腺病毒为载体将TGF-β1基因转染至髓核细胞内,RT-PCR和免疫组化方法检测TGF-β1的表达,并检测其对髓核细胞基质中蛋白多糖含量的影响。结果兔髓核细胞贴壁及生长的速度均十分缓慢,Ad-TGF-β1转染的髓核细胞内可检测到TGF-β1基因的表达,并且可持续到8周,Ad-TGF-β1转染后的髓核细胞合成蛋白多糖的量增加。结论腺病毒介导的TGF-β1基因可以在髓核细胞内得到持续的表达,并且产生明显的生物学效应。 Objective To detect the expression of transforming growth factor β1 ( TGF-β1 ) gene mediated by adenovirus in rabbit nucleus pulposus cells and its biological effects on proteoglycans. Methods The rabbit nucleus pulposus cells were isolated and cultured in vitro. After transduced into the rabbit nucleus pulposus cells mediated by adenovirus, the expression of TGF-β1 gene was evaluated with RT-PCR and immunohistochemical methods and its biological effects on proteoglycans was evaluated. Results The amount of nucleus pulposus cells cultured in vitro increased slowly. The expression of TGF-β1 gene could be detected in the nucleus pulposus cells transfected with Ad-TGF-β1, and persisted 8 weeks. The content of proteoglycans synthesized by nucleus pulposus cells increased when tansfected with Ad-TGF-β1. Conclusion The TGF-β1 gene mediated by adenovirus could persistently express in nucleus pulposus cells, and produce biological effects on nucleus pulposus cells.
