摘要
介绍一种分离、鉴定肝实质细胞的新方法.采用恒流泵、不锈钢滤网等简易灌流装置,对成年大鼠进行原位灌注分离肝实质细胞;并根据肝实质细胞能组成型表达6-磷酸葡萄糖酶这一特性,采用酶细胞化学技术鉴定肝实质细胞.经本实验改进的肝实质细胞分离方法具有简便、快捷、可靠的特点,为进一步研究肝实质细胞的各种功能奠定了基础.
A new method was introduced to isolate and identify rat hepatocytes in this paper. The rat liver was perfused with the solution of collagenase to obtain liver cells suspension. The simple perfusing device consisted of squirmy pump and sieve and so on. The purity of rat hepatoeytes were identified by the constituted expression of 6-P-Gase using enzyme-chemistry. The method is simple, convenient and reliable for isolating and identifying the hepatocytes of rat and found a basis for further research of hepatocytes.
