葛根素对实验性大鼠肺纤维化影响 被引量:17

摘要 目的观察葛根素对肺纤维化大鼠的影响。方法大鼠随机分为对照组、模型组、葛根素组,以博来霉素气管内滴入建立大鼠的肺纤维化模型,第29天取肺组织作病理切片及HE染色和胶原纤维染色,以免疫组化法(SP法)检测I型胶原纤维,以RT-PCR法检测TGFβ1,并进行图像分析定量,进行统计分析。结果葛根素组相比模型组在总胶原纤维、I型胶原纤维和TGFβ1mRNA的表达均有下降。结论葛根素可抑制炎症和成纤维细胞增殖,抑制TGFβ1mRNA的表达,并可减少肺胶原纤维的形成,起到抑制和延缓实验大鼠肺纤维化的作用。 Objective To investigate the effects of Kakkonein on pulmonary fibrosis of rats in lab research were divided into three groups: controls, untreated and Kakkonein groups. HE stain, collagen stain, RT-PCT and Methods The rats methods were applied to rats' lungs on the 29th days after the beginning of the experiment. The statistical analysis was performed on the data. Results There was more decreased value of total collagen, Type Ⅰ collagen and TGFβ1 mRNA in Kakkonein group than in the untreated animal groups. Conclusion Kakkonein can restrain the formation of pulmonary fibrosis by decreasing inflammation, fibroblast proliferation, Type Ⅰ collagen formation and gene expression of TGFβ1.
